Proteasome activity contributes to pro-survival response upon mild mitochondrial stress in Caenorhabditis elegans

Defects in mitochondrial function activate compensatory responses the cell. Mitochondrial stress that is caused by unfolded proteins inside organelle induces a transcriptional response (termed “mitochondrial protein response” [UPRmt]) mediated activating transcription factor associated with 1 (ATFS-1). The UPRmt increases quality control. dysfunction frequently causes defects import of proteins, resulting accumulation outside organelle. In yeast, cells respond to mistargeted increasing activity proteasome cytosol “unfolded activated mistargeting proteins” [UPRam]). presence and relevance this higher eukaryotes unclear. Here, we demonstrate Caenorhabditis elegans lead activation life span extension. Both prolongation partially depend on ATFS-1, despite its lack influence proteasomal gene transcription. Importantly, depends fully assembled proteasome. Our data provide link between direct mechanisms promote longevity.
